Mycoplasma,thesmallestandself-replicatingprokaryotes,arecommoncontaminantsofprimarycellsandcelllines.Mycoplasmacontaminationcanaffectcellgrowth,morphologyandmetabolism.Mycoplasmaareabletopassthrough0.2µmfiltersduetotheirsmallsize.FurThermore,unlikebacteriaandfungi,mycoplasmacontaminationisresistanttocommonlyusedantibiotics,andcannotbeidentifiedvisually.Duetothedifficultiesindetectingmycoplasma,cellsshouldberegularlyscreenedtocontrolmycoplasmacontamination.ConventionalscreeningmethodsincludemicroBIOLOGicalculture,fluorescentDNAstaining,andbiochemicaldetectionmethods,andaretypicallytimeconsumingandlacksensitivity.Asaresult,polymerasechainreaction(PCR)basedmethodshavebeendevelopedasaquickandconvenientdetectionoflowlevelmycoplasmainfection,whileprovidinghighsensitivityandspecificity.
Alignmentstudiesofmycoplasma16SrRNAsequencesrevealedtheexistenceofhighlyconservedsequences.PrimerstargetingthesesequencesallowforthespecificamplificationofthemycoplasmaDNAfragment.ScienCell™MycoplasmaPCRDetectionKitisa16SrRNA-basedPCRassayconsistingof2×PCRmastermix(whichincludesPCRbuffer,DNApolymeraseanddNTPs),mycoplasmaprimerset,andapositivecontrol,whichisnoninfectiousgenomicDNA(gDNA)of M.fermentans.Thegenus-specificprimersetrecognizesthefivetypicalcontaminatingmycoplasmaspecies(i.e.,M.hyorhinis,M.arginini,M.orale,M.fermentans,andA.laidlawi),whichaccountfor98%ofcontamination,aswellasmembersofthegeneraUreaplasma,Spiroplasma,andAcholeplasma,whichaccountfortheremaining2%ofcontamination.EukaryoticandbacterialDNAisnotamplifiedbythiskit.Thesizeoftheexpectedbandis280bp.
