bFGF-StimulatedMouseEmbryonicFibroblast-ConditionedMedium(bFGF-stdMEF-cm)isacompleteserum-freemediumforthefeeder-freecultureofhumanpluripotentstemcells(hPSCs),includingembryonicstemcellsandinducedpluripotentstemcells.bFGF-stdMEF-cmisacompletemediumcontainingrecombinanthumanbasicFGF(rhbFGF)andisreadytouseafterthawing,sothereisnoneedtoaddextrabasicFGF.Itsupportstheself-renewalofhPSCandmaintainspluripotencyinthepresenceofhPSCqualifiedmatrix(e.g.,BDMatrigelTMmatrix).
bFGF-stdMEF-cmwaspreparedusinghighqualitymitomycinCtreatedMEF(MEF-mt,Cat.#M7570-mt)derivedfromE13embryosofCF1mice.SerumfreemediumwasconditionedwithmitomycinCtreatedCF1MEFsfor24hours.Theconditionedmediumwascollecteddailyfor7days,pooledandsterilizedbyfilteringthrougha0.22µmfilter.RecombinanthumanbasicFGF(rhbFGF,Cat.#104-02)wasaddedtothemediumatafinalconcentrationof4ng/mLtomakethecompletemedium.
bFGF-stdMEF-cmhasbeentestedforitsABIlitytosupporthPSCself-renewalonBDMatrigelTMmatrixinvitro.Themediumcolorofeachlotmayvarybutdoesnotaffectperformance.
